NNMTi is an enzyme overexpressed during skeletal muscle aging and is associated with the repair of damage to the NAD+ pathway, dysregulation of sirtuin1 activity, and increased senescence of muscle stem cells.
NNMTi is a potent inhibitor of nicotinamide N-methyltransferase (NNMT) (IC50=1.2μM) and can selectively bind to NNMT substrate binding site residues. NNMTi can promote myoblast differentiation in vitro and enhance the fusion and regeneration capacity of muscle stem cells in aged mice.
